技术领域
:本发明涉及一类1,2-二苯基乙垸类尿素酶抑制剂的制法以及它们在制备抗胃炎、胃溃疡药物中的应用。
背景技术
:幽门螺旋杆菌(说//^""^^/^/)会引发胃炎、胃溃疡、十二指肠溃疡、胃萎缩、肠上皮化生、胃癌、胃淋巴瘤等多种疾病。1994年世界卫生组织和国际癌症研究中心将H;^/or/列为第一类致癌因子。据统计,大约世界人口的一半感染上了H;^/on',在发展中国家中感染率高达80-90%。我国的感染率为60%左右。胃炎患者的/Z;^/on'检出率为80-卯%,消化性溃疡患者更高,达95%以上。超过卯%的十二指肠溃疡和80%左右的胃溃疡是Hp^on'所致。治疗上述疾病以及防止复发的前提是先根除Hj^/w/。目前根除H户;;/w/最常用的是三联法一种质子泵抑制剂(奥美拉唑或兰索拉唑)和两种抗生素(阿莫西林、氧氟沙星或甲硝唑)。但是,奥美拉唑有明显的副作用除会引起腹痛、呕吐、胀气等副作用外,还会引起肝重量增大等;还有诱发胃类癌、引起肾衰等危险,而且H;^/oW对所用的抗生素容易产生耐药性,这一方法的有效率正逐年下降。幽门螺旋杆菌能在强酸性环境的人胃里存活的最主要的原因是它能产生尿素酶,尿素酶水解尿素释放出来的氨能提高pH值,因而为/Z.;^/w7'营造了一个适宜的微环境。其他一些病菌,如普通变形杆菌(iVofewsvw/gaWs)、奇异变形杆菌(Pratowm/raWto)、解脲脲原体(Oe叩/a训awm^"c)等,当它们感染尿路系统后,因为尿素酶的作用引起尿的pH升高,导致磷酸铵镁等物质的沉淀,进而发展成尿路结石。具有尿素酶活性的病原菌要么靠尿素酶水解尿素产生氨为自身的生命活动提供氮源,要么利用氨的碱性为其生存提供一个适宜的微环境。故阻断了尿素酶活性,就能有效的杀灭这类病菌。目前,尿素酶抑制剂已成为治疗这类疾病的一线药物。但现有的尿素酶抑制剂存在一些不足,比如羟氨酸类具有致(大鼠)畸变性,磷酸二酰胺类在酸性环境中不稳定。因此新型高效低毒尿素酶抑制剂的筛选是开发这类药物的关键。2003年,Matsubara等人报道表没食子儿茶素(I)具有很好的尿素酶抑制活性。本发明通过对化合物I的改造获得了具有II所示的结构的一系列l,2-二苯基乙垸类尿素酶抑制剂。试验表明,有些化合物对尿素酶表现出较好的抑制活性。
